Labeling type (Brightfield or Fluorescent)

By default, we utilize an HRP-catalyzed DAB system for visualization under brightfield. Please specify if you need fluorescent labeling and your preferred excitation wavelength (we will choose if left blank), or if you need multiplex brightfield staining.

*Note: we cannot scan fluorescently labeled slides. We have a scope (90i) equipped with a camera and Nikon elements software that you may schedule time on to manually capture images if desired.

Isotype control sample*

Please specify here which sample ID should be used as the isotype control so we may cut an extra slide. Please consult with us if you are unsure what makes an appropriate isotype control. Typically, the same species, tissue, and gene treatment should suffice. An isotype control is incubated with an antibody of irrelevant target(s) that shares the same heavy and light chain as the target antibody. Isotype controls ensure the observed staining is due to specific antibody binding to the target rather than an artifact.

Special Instructions

We utilize a Hamamatsu Nanozoomer automated slide scanning instrument with an auto-focusing feature. Uploading a diagram can assist us to select manual focal points if needed. This instrument does not scan fluorescently labeled slides at this time.

Image Analysis with Visiopharm

Instructions and/or needs

Visiopharm is equipped with standard binary and region-of-interest tools for quantification as well as machine learning and AI functionalities for more involved studies. Our typical analysis consist of generating a ratio of positive stain to total tissue area. Please describe the measurements you're interested in so the image analyst can make suggestions and develop an appropriate APP for your study. Advanced identification and quantification methodologies will require a consult with the image analyst.

In-Situ Hybridization (ISH)

ISH has multiple prerequisites to be successful. Email bfount@uw.edu for consultation before fixing your tissue, if possible.
The investigator must purchase the probe from ACDBio and verify with the manufacturer that it is compatible with the Leica Bond Rx. We supply the positive, negative, and ISH kits for use on our Leica Bond Rx. Email us if you need assistance acquiring a compatible probe.
